Numerous studies have explored plant-derived Extracellular Anti-melanogenic as a natural alternative to chemical-based agents, without the side effects. Our recent research examined the anti-melanogenesis properties of extracellular vesicles (EVs) extracted from the leaves and stems of S. japonica. Through spectrophotometric and biochemical techniques, we discovered that both leaf-derived extracellular vesicles (LEVs) and stem-derived extracellular vesicles (SEVs) effectively reduced melanin content in a concentration-dependent manner in the B16BL6 mouse melanoma cell line, along with decreased Tyrosinase (TYR) activity.
Extensive analysis and research confirmed that both LEVs and SEVs led to a concentration-dependent reduction of melanin content in melanoma cells. In comparison to arbutin, a commonly used positive control, both LEVs and SEVs exhibited a superior whitening effect on melanoma cells, with LEVs being particularly effective.
Importantly, neither LEVs nor SEVs exhibited significant cytotoxicity. We also investigated the impact of plant-derived extracellular vesicles on the expression of tyrosinase-related proteins (TRPs) in melanoma cells. LEVs effectively inhibited the expression of melanogenesis-related genes and proteins, including microphthalmia-related transcription factor (MITF), TYR, TRP-1, and TRP-2. In a human epidermal model, LEVs demonstrated a more potent inhibitory effect on melanin production compared to arbutin.
In conclusion, our findings indicate that LEVs derived from S. japonica present a promising natural anti-melanogenic agent for use in medicinal formulations, providing a new avenue for enhancing skin radiance and rejuvenation.
